چکیده

The quantum yield, under non-photoreactivating conditions, for inactivation of tobacco mosaic virus RNA by ultraviolet radiation (254 nm.) is independent of the RNA concentration but varies appreciably with the ionic strength of the irradiation solvent, being approximately three times greater in water than in o-I M-potassium phosphate buffer. Photoreactivation of inactivated tobacco mosaic virus RNA is dependent on RNA concentration, ionic strength of solutions during irradiation, and on the light quality used for photoreactivation. These data can be interpreted as evidence that pyrimidine hydrate is a lethal lesion at least in RNA irradiated at low ionic strength.

The Ultraviolet Light and Photosensitized Inactivation of Tobacco Mosaic Virus

The quantum yield for the inactivation of tobacco mosaic virus has been determined at 253.7 mmicro and found to be 4.3 x 10(-6). The possible significance of the observed one-hit process of inactivation has been discussed in terms of the kinetics and the rupture of model substances including nucleic acid. The ultraviolet light inactivation, which proceeds independent of oxygen, occurs without c...

Dark reactivation of ultraviolet-irradiated tobacco necrosis virus.

Some damage caused in tobacco necrosis virus by u.v. radiation could be repaired in darkness in Chenopodium amaranticolor (dark reactivation) but not in French bean or in tobacco. By contrast, photoreactivation of the irradiated virus was observed in French bean and in tobacco but not in Chenopodium. The kind of damage in u.v.-irradiated virus that is susceptible to repair by photoreactivation ...

Studies on the ability of light to counteract the inactivating action of ultraviolet radiation on plant viruses.

Of seven plant viruses tested, all except tobacco mosaic showed the phenomenon of photoreactivation, i.e. plants exposed to visible light after inoculation with preparations partially inactivated by ultraviolet radiation produced more local lesions than plants kept in darkness. Five straiIis of tobacco mosaic virus, which differed widely in their pathogenicity and other properties, were tested,...

The Reversible Inactivation of Tobacco Mosaic Virus by Crystalline Ribonuclease

The reversible inactivation of tobacco mosaic virus by crystalline ribonuclease is reported. Studies on the effect of time of standing on the amount of inactivation, and on the effect of dilution and repeated high speed centrifugation on the recovery of virus activity, and the preparation of an insoluble virus-enzyme complex show that the inactivation is brought about at least in part by a comb...

Visualization and characterization of tobacco mosaic virus movement protein binding to single-stranded nucleic acids.

Cell-to-cell spread of tobacco mosaic virus (TMV) is presumed to occur through plant intercellular connections, the plasmodesmata. Viral movement is an active process mediated by a specific virus-encoded P30 protein. P30 has at least two functions, to cooperatively bind single-stranded nucleic acids and to increase plasmodesmatal permeability. Here, we visualized P30 complexes with single-stran...
